Lab Services
DNA Software employs leading experts in the thermodynamics and hybridization of nucleic acids and provides laboratory and consulting services to major biotech companies working on development of nucleic acid based assays and therapies, with one major feature being DNA and RNA melting curve analyses.
DNA & RNA Melting Curve Analysis:
-
Our laboratory has state-of-the art equipment and procedures to perform UV thermal denaturation studies of nucleic acids (DNA, RNA and modified oligonucleotides.
-
In the past three years we delivered more than a dozen contract projects and determined thermodynamic parameters for hundreds of oligonucleotide duplexes, hairpins, and single and triplex oligonucleotide systems.
-
Assay Development:
-
Effect of probe modification such as fluorophore incorporation or nucleoside modifications on probe hybridization stability.
-
Effect of probe modification on probe specificity and SNP discrimination efficiency.
-
Effect of buffer conditions and additives on hybridization stability.
-
Consulting on assay design strategy and experimental testing
Custom Probe Design:
Formulation and Manufacturing:
Regulatory Submissions:
-
Characterization of duplex/hairpin thermodynamics: melting temperature, dG, dH, and dS, and secondary structure.
Wet Lab Contract Research:
-
Large scale and small scale projects to determine the thermodynamics of modified nucleotides. In the past we have investigated: PNA, LNA, inosine, MGB, DMSO, formamide, TMAC, fluorophores and many other modifieds and incorporated the parameters into software.
-
Deep expertise in the design of oligonucleotides for the determination of physical parameters for DNA and RNA thermodynamics.
-
DNA Software has had six NIH funded phaseII SBIR grants and one contract from the Department of Homeland Security to continue to push the field forward.
